Overview

On 9/23 we did our final assessment of the Sequim environmental hardening oysters for Effort A and closed out this project! We checked on the PolyIC outplants and left those deployed.

Final assessment

We did the following today to assess and close out the Effort A experiment:

  • Collected all Effort A seed bags (7 bags) from the lines and brought them up to the beach
  • Removed oysters from each bag and counted mortalities
  • Arranged oysters for a photograph to capture number of live animals and for growth analysis
  • Tag and scale bar included in each photo
  • Subsampled oysters from each bag to total n=~30 oysters per treatment group (2 treatment groups total) into red mesh bags with their own tag labels for survival assays in the lab (see below)
  • Bags and equipment were cleaned
  • Two temperature loggers were downloaded that were brought in on the bags. There is still one logger remaining with the PolyIC outplants in the field and I added one more to an outplant bag.
  • I recorded data and uploaded size image photos.

All data is in our 10K seed GitHub repo.

Samples for survival assays

All data is in our 10K seed GitHub repo in the lab survival folder.

Here is a table of the tags collected today from Sequim. I will use this to help Jesse and Maddy prepare datasheets for assays. Each bag has about 30-35 oysters. 30 will be used for collections.

See our methods plan in my previous post here for the survival assays.

tag experiment location treatment
49 Effort A Sequim Treated
50 Effort A Sequim Control

PolyIC checks

The PolyIC oysters look good! All 8 bags are still deployed.

To do

  • Start final photo analysis for growth and survival
  • Survival assays in the lab (Jesse & Maddy)
